| AnexinA5isaphospholipidbindingprotein,whichbindswithhighaffinityandselectivitytoPSinthepresenceofcalcium.PSispredominantlylocatedinmembraneleaflets,whichfacethecytosol.However,recentfindingsshowthateachcelltypehasthemolecularmachinerytoexposePSatitscellsurface.Thismachineryisactivatedduringtheexecutionofapoptosis.OncePSisexposedatthecellsurfaceitexhibitsprocoagulantandproinflammatoryactivities.AnexinA5willbindtothePS-exposingapoptoticcellandcaninhibittheprocoagulantandproinflammatoryactivitiesofthedyingcell.AnexinA5hasalsobeenidentifiedasananticoagulantproteininthebloodcoagulationcascade,byactingasaninhibitorofprothrombinactivation.ThepresenceofantibodiestoAnexinA5isassociatedwithsystemiclupuserythematosus(SLE),recurrentspontaneousabortionsandsystemicsclerosis(SSc). |
