| BACKGROUND: | TheproteinencodedbythisgeneisamemberofthesuperfamilyofATP-bindingcassette(ABC)transporters.ABCproteinstransportvariousmoleculesacrossextra-andintra-cellularmembranes.ABCgenesaredividedintosevendistinctsubfamilies(ABC1,MDR/TAP,MRP,ALD,OABP,GCN20,White).ThisproteinisamemberoftheALDsubfamily,whichisinvolvedinperoxisomalimportoffattyacidsand/orfattyacyl-CoAsintheorganelle.AllknownperoxisomalABCtransportersarehalftransporterswhichrequireapartnerhalftransportermoleculetoformafunctionalhomodimericorheterodimerictransporter.Thefunctionofthisperoxisomalmembraneproteinisunknown;howeverthisproteinisspeculatedtofunctionasadimerizationpartnerofABCD1and/orotherperoxisomalABCtransporters.Mutationsinthisgenehavebeenobservedinpatientswithadrenoleukodystrophy,aseveredemyelinatingdisease.Thisgenehasbeenidentifiedasacandidateforamodifiergene,accountingfortheextremevariationamongadrenoleukodystrophyphenotypes.ThisgeneisalsoacandidateforacomplementgroupofZellwegersyndrome,ageneticallyheterogeneousdisorderofperoxisomalbiogenesis. |
