| ANGPTL4(Angiopoietin-likeprotein4)mainlyexpressedinendothelialcells(hypoxia-induced).RegulatesangiogenesisandmodulatestumOrigenesisanddirectlyregulateslipid,glucose,andenergymetabolism.Inhibitsproliferation,migration,andtubuleformationofendothelialcellsandreducesvascularleakage.ANGPTL4isaproteinconsistingofanN-terminalcoiled-coildomainandaC-terminalfibrinogen-likedomain(FLD).Bothdomainshavedistinctbiologicalfunctions.Thecoiled-coildomainisresponsIBLefortheinhibitoryeffectsonlipoproteinlipase(LPL)convertingtheactiveformofLPLintoaninactiveform,andtheFLDdomainmediatesitsantiangiogenicfunctions.ThecoiledcoilandtheFLDdomainsareseparatedbyashortlinkerthatcanbecleavedaftersecretion.ANGPTL4appearsonthecellsurfaceasthefull-lengthform,whereitcanbereleasedbyheparintreatment.ANGPTL4proteinisthenproteolyticallycleavedbyproproteinconvertases(PCs),includingfurin,PC5/6,pairedbasicaminoacid-cleavingenzyme4,andPC7. |
