Background
| Angiopoietin-2isalsoknownasANGPT2,AGPT2,ANG2,andisasecretedglycoproteinthatplaysacomplexroleinangiogenesisandinflammation.Ang2iswidelyexpressedduringdevelopment,butitisrestrictedpostnatallytohighlyangiogenictissuessuchastheplacenta,ovaries,anduterus.Itisparticularlyabundantinvascularendothelialcells(EC)whereitisstoredinintracellularWeibelPaladebodies.BothAng2andtherelatedAngiopoietin1(Ang1)areligandsforthereceptortyrosinekinaseTie2.Ang2functionsasaproangiogenicfactor,althoughitcanalsoinduceECdeathandvesselregression.UponitsreleasefromquiescentEC,itregulatesvascularremodelingbypromotingECsurvival,proliferation,andmigrationanddestABIlizingtheinteractionbetweenECandperivascularcells.Ang2isrequiredforpostnatalvascularremodeling,anditcooperateswithAng1duringlymphaticvesseldevelopment.ItmediatestheupregulationofICAM1andVCAM1onEC,whichfacilitatestheadhesionofleukocytesduringinflammation.Ang2competitivelyinhibitAng1-inducedendothelialcellresponsesmediatedbyTie2,andreducesvascularintegrity.ButtheroleofAng2iscontroversialsincetheoppositeoutcomeshasbeenreportedinotherstudies.Over-expressionofAng2disruptsthevascularremodeling,induceendothelialcellapoptosis,andmayplayanimportantregulatingroleintumorangiogenesis.Ang2alsopromotestheneuronaldifferentiationandmigrationofsubventricularzoneProgenitorcells. |
