| BACKGROUND: | 14-3-3proteinsareafamilyofhighlyconservedproteinsthatappeartohavemultiplerolesincellsignaling(BridgesandMoorhead,2005).Theproteinsareabundantlyexpressedinthebrainandhavebeendetectedinthecerebrospinalfluidofpatientswithdifferentneurologicaldisorders(Bergetal.,2003).14-3-3proteinsbindproteinligandsthataretypicallyphosphorylatedonserineorthreonineresiduesandregulatethefunctionsofthesebindingpartnersbyanumberofdifferentmechanisms(Silhanetal.,2004;DoughertyandMorrison,2004).The14-3-3proteinsaffectadiversearrayofcellularprocessesincludingthecellcycleandtranscription,signaltransductionandintracellulartrafficking.Thesefunctionsof14-3-3proteinsarefacilitatedby,ifnotdependenton,itsdimericstructure.Recentworkhasdemonstratedthatthedimericstatusofthe14-3-3proteinisregulatedbysite-specificserinephosphorylation(Woodcocketal.,2003). |
